华美奥锹甲不同颚型雄性成虫肠道细菌群落多样性

Diversity of gut bacterial communities in male adults of Odontolabis fallaciosa (Coleoptera: Scarabaeoidea: Lucanidae) with different mandibular forms

摘要: 【目的】分析比较华美奥锹甲Odontolabis fallaciosa不同颚型雄性成虫肠道细菌的群落结构和多样性。【方法】分别提取华美奥锹甲雄性3种颚型(大颚型、中颚型和小颚型)31头成虫样本(15个大颚型、10个中颚型、6个小颚型)的肠道DNA,利用Illumina MiSeq技术对其肠道细菌的16S rDNA V3-V4变异区序列进行测序,统计样品操作分类单元(OTU)数量,分析物种组成丰度、α多样性和β多样性。【结果】共获得优质序列2 238 637条,在97%相似度下可将其聚类为用于物种分类的3 256 个OTU,共注释到的主要分类阶元有42个门、328个科和542个属。门分类水平上,变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)、厚壁菌门(Firmicutes)、柔膜菌门(Tenericutes)为优势门类;属分类水平上,优势属为不动杆菌属AcinetobacterDysgonomonas属、巴尔通氏体属Bartonella、金黄杆菌属Chryseobacterium。α多样性分析结果表明,华美奥锹甲雄性成虫肠道细菌群落整体多样性比较丰富;β多样性分析结果表明,大颚型、中颚型和小颚型之间OTU组成有差异。【结论】采用Illumina MiSeq测序技术,分析了华美奥锹甲雄性成虫肠道细菌群落的结构和组成。大颚型与中颚型雄性成虫间的肠道细菌群落组成差异显著,但大颚型与小颚型间、中颚型与小颚型间雄性成虫肠道细菌群落组成无差异。

Abstract: 【Aim】 To analyze and compare the community structure and diversity of the gut bacteria in adult males of Odontolabis fallaciosa with different mandibular forms. 【Methods】 The total DNA of guts from 31 male adults of O. fallaciosa with three mandibular forms (large, medium and small mandible forms) (including 15 adult males of large mandible form, 10 adult males of medium mandible form and 6 adult males of small mandible form) were extracted. The 16S rDNA gene fragments (V3-V4 region) of gut bacteria were sequenced by Illumina MiSeq, and the number of operational taxonomic units (OTUs), species composition, abundance, and alpha and beta diversity were analyzed. 【Results】 A total of 2 238 637 high quality sequences were obtained and clustered into 3 256 OTUs at a 97% similarity threshold. Analysis showed that they could be annotated into 42 phyla, 328 families and 542 genera. Among them, four phyla (Proteobacteria, Bacteroidetes, Firmicutes and Tenericutes) were prevalent in the gut bacteria communities of male adults, and four genera (Acinetobacter, Dysgonomonas, Bartonella and Chryseobacterium) were dominant. The alpha diversity analysis showed that the gut bacteria were quite abundant in these male samples of O. fallaciosa, and the beta diversity revealed that the composition of OTUs was different among the large, medium and small mandible males. 【Conclusion】 Using Illumina MiSeq sequencing technology, the structure and composition of the gut bacterial community in adult males of O. fallaciosa were analyzed. The microbial community composition in guts of adult males of O. fallaciosa has significant difference between the large and medium mandible adult males, whereas no significant difference exists between the large and small mandible adult males, or between the medium and small mandible adult males.
