小菜蛾乙酰胆碱酯酶cDNA片段的克隆和序列分析

摘要: 利用反转录-多聚酶链式反应(RT-PCR)的方法对小菜蛾Plutella xylostella的乙酰胆碱酯酶基因cDNA片段进行了克隆和序列分析。通过简并性上游引物和下游引物扩增出了小菜蛾乙酰胆碱酯酶基因281bp的cDNA片段。同源性分析表明, 该cDNA片段与其它昆虫乙酰胆碱酯酶基因序列具有较高的同源性。

Abstract: Using RT-PCR, the partial cDNA sequence of the acetylcholinesterase gene in the diamondback moth, Plutella xylostella (L.) was analyzed. With the use of a pair of degenerate primers, one fragment of 281 bp was amplified from the fourth instar larva. Homologous analysis of the cloned cDNA amino acid sequence of acetylcholinesterase in diamondback moths and corresponding sequences of other source of amino acid sequences of acetylcholinesterase revealed that there is high degree of amino acid sequence homology between the diamondback moth and other insects.
