B型烟粉虱抗噻虫嗪品系的遗传分化

Genetic differentiation in the thiamethoxam-resistant strain of Bemisia tabaci B-biotype

摘要: 采用扩增片段长度多态性(Amplified fragment len gth polymorphism, AFLP )技术, 研究了B型烟粉虱Bemisia tabaci 噻虫嗪 (thiamethoxam ) 抗性品系在DNA分子水平上的遗传分化。实验数据用STATISTICA (4.5版本) 软件进行分析, 遗传距离用UPGMA (非加权对组算术平均值聚类) 方法进行聚类。在聚类树中, 首先是抗性个体与敏感个体聚为2枝, 然后雌、雄个体聚为2枝, 说明抗性品系在DNA分子水平上已存在明显分化。另外, 利用Eco RⅠ+ACT/MseⅠ+CTG引物, 分别在全部抗性个体中扩增出了敏感个体所没有的特异性同一片段和在全部敏感个体中扩增出了抗性个体所没有的特异性同一片段, 经克隆、测序, 这2个片段分别是150 bp 和315 bp。此2个片段, 可作为烟粉虱抗噻虫嗪品系和敏感品系的分子标记而在抗性监测中得以应用。

Abstract: The genetic differentiation in the thiamethoxam-resistant strain of the B-biotype Bemisia tabaci was investigated by the method of AFLP marker. Then a dendrogram was constructed from the matrix of genetic distances using the STATISTICA software (4.5 version) and UPGMA (Unweighte d Pair-Group Method using the Arithmetic Averages). A significant genetic differentiation was found between the resistant and susceptive strains of Bemisia tabaci. Additionally, characteristic amplified DNA fragments, the fragment 150 bp for resistant strain and the fragment 315 bp for susceptive strain were identified using the EcoRⅠ+ACT/RMseRⅠ+CTG primer, which may serve to develop molecular detection of Bemisia tabaci resistance to thiamethoxam.
