绿盲蝽酯酶基因AlucEST1的克隆及表达谱分析

摘要: 昆虫体内的气味降解酶(odorant degrading enzymes, ODEs)能够降解气味分子, 使嗅觉受体免受持续的刺激。酯酶作为气味降解酶类的一种, 在昆虫嗅觉行为过程中发挥重要作用。本研究利用RACE技术克隆了一个新的绿盲蝽Apolygus lucorum酯酶基因AlucEST1 (GenBank登录号: JQ715614)。序列分析结果表明, 该基因全长1 810 bp, 开放阅读框为1 713 bp, 编码571个氨基酸。N-末端疏水区包含由起始位置开始的一段17个氨基酸的信号肽。通过半定量RT-PCR和Real-time qPCR 技术解析该酯酶基因在不同组织的表达谱。结果发现, 该酯酶基因在绿盲蝽的触角、 头、 胸、 腹、 足、 翅等部位中均有表达。其中, AlucEST1在胸、腹部中表达量最高, 在触角中也大量表达。据此推测该基因在气味分子的降解中起重要作用, 同时也参与绿盲蝽体内的多种代谢调控。

Abstract: Odorant degrading enzymes (ODEs) can degrade the excess odor molecules so as to prevent olfactory receptors from the sustained stimulation. Esterase is a kind of odorant degrading enzyme, and plays an important role in the process of insect olfactory behavior. In this study, a novel putative ODE gene AlucEST1 (GenBank accession number: JQ715614) from the green plant bug, Apolygus lucorum was cloned by using rapid-amplification of cDNA ends (RACE) technique. Sequence analysis showed that AlucEST1 is 1 810 bp in length with the open reading frame of 1 713 bp, and encodes 571 amino acids. The N-terminal hydrophobic region contains a 17 amino acid signal peptide. Semi-quantitative RT-PCR and Real-time qPCR were conducted to explore the AlucEST1 expression in different tissues of A. lucorum. The results showed that AlucEST1 was expressed in various tissues including antennae, head, thorax, abdomen, legs and wings. AlucEST1 had the highest expression level in thorax and abdomen, and was also abundantly expressed in the antennae. It is so inferred that the gene plays an important role in the degradation of odor molecules and is also involved in a variety of metabolic regulation in A. lucorum.
