Studies on genetic variations and phylogenetic relationships among five species of Tetrix using RAPD markers

Abstract: Random amplified polymorphic DNA(RAPD)techniques were applied to assess genetic variations among five species in the genus Tetrix. Amplifications with 12 primers under predetermined optimal reaction conditions gave 84 reproducible fragments ranging from 200 to 2 000 bp.The amplified products were scored as present (1) or absent (0) for each DNA sample and an index of genetic similarity (F) was calculated. The value of D was used to evaluate genetic distances between species and a phylogenetic tree was constructed.
