虫龄对杜仲梦尼夜蛾幼虫肠道细菌组成和多样性的影响

Effect of larval instars on the gut bacterial composition and diversity of Orthosia songi (Lepidoptera: Noctuidae)

摘要: 【目的】杜仲梦尼夜蛾Orthosia songi是危害杜仲Eucommia ulmoides叶片的单食性害虫。本研究旨在探究不同龄期杜仲梦尼夜蛾幼虫肠道菌群的特征。【方法】使用Illumina HiSeq高通量测序技术调查杜仲原产地陕西省略阳和河南省灵宝杜仲梦尼夜蛾1, 3和6龄幼虫肠道细菌组成和多样性。【结果】两个地区杜仲梦尼夜蛾幼虫肠道细菌群落在门水平均以假单胞菌门(Pseudomonadota)、蓝菌门(Cyanobacteriota)、放线菌门(Actinomycetota)、芽孢菌门(Bacillota)和拟杆菌门(Bacteroidota)为优势菌门,各个菌门的丰度在不同龄期样品间不同,并且随着杜仲梦尼夜蛾幼虫龄期的增大假单胞菌门的丰度逐渐减少,芽孢菌门的丰度逐渐增大。在属水平,不动杆菌属Acinetobacter为优势菌群,其丰度随着杜仲梦尼夜蛾幼虫龄期的增大而减小。多样性分析结果显示,杜仲梦尼夜蛾不同龄期幼虫样品之间肠道细菌α多样性指数(Shannon指数、 Simpson指数、 ACE指数和Chao指数)差异不显著,β多样性存在显著差异。【结论】本研究结果说明陕西省略阳和河南省灵宝杜仲梦尼夜蛾幼虫肠道细菌菌群组成相似,但不同龄期幼虫之间肠道细菌菌群组成存在显著差异,提示杜仲梦尼夜蛾幼虫肠道微生物种群受到该虫生长发育阶段的影响。

Abstract: 【Aim】Orthosia songi is a monophagous pest damaging Eucommia ulmoides leaves. The objective of this research is to explore the characteristics of the gut bacterial communities of O. songi larvae at different instars.【Methods】 Illumina HiSeq high-throughput sequencing was employed to investigate the composition and diversity of the gut bacteria of the 1st, 3rd and 6th instar larvae of O. songi from Lüeyang, Shaanxi Province and Lingbao, Henan Province. 【Results】 At the phylum level, the gut bacterial communities of O. songi larvae from the two regions were dominated by Pseudomonadota, Cyanobacteriota, Actinomycetota, Bacillota and Bacteroidota, and the abundance of various phyla differed between samples of larvae at different instars. Notably, the abundance of Pseudomonadota decreased and that of Bacillota increased gradually with the increase of the larval instars of O. songi. At the genus level, Acinetobacter was the dominant bacterium, and its abundance also decreased with the increase of the larval instars of O. songi. Diversity analysis result showed that there were no significant differences in the gut bacterial alpha diversity indexes (Shannon index, Simpson index, ACE index and Chao index) among samples of O. songi larvae at different instars, while the beta diversity of the gut bacterial diversity among samples of O. songi larvae at different instars showed significant difference. 【Conclusion】 The results of this study suggest that the gut bacterial community composition of O. songi larvae collected from Lüeyang, Shaanxi Province and Lingbao, Henan Province is similar, but there is significant difference in the gut bacterial community composition among different larval instars, suggesting that the gut microbiota of O. songi larvae is influenced by their growth and developmental stages.
